Research Topic: Impact of 19, 20-Epoxydocosapentanoic Acid on the Production of Matrix Metalloproteinases by Pro-Fibrotic Monocytes Graduation Year: 2025

Undergraduate Research in the Neighborhoods

Prior studies have demonstrated that 19, 20-Epoxydocosapentaenoic acid (19,20-EDP) modifies the phenotype of myeloid cells such as monocytes. However, the mechanism is unknown. The main goal of Ladymar’s project was to test the hypothesis of 19,20-EDP stimulating the conversion of monocytes into MMP-producing macrophages. Monocytes were purified from the livers of mice with fibrosis and treated with 19,20-EDP. mRNA levels of Matrix-Metalloproteinases (MMPs) and proinflammatory cytokines were quantifies by real-time PCR. Results showed that 19,20-EDP produced a change in cell morphology consistent with the conversion into macrophages. 19,20-EDP treatment increased mRNA levels of MMP-9 and MMP-13 while decreasing levels of tumor necrosis factor-alpha (TNF-a) and interleukin-6 (IL-6). It was concluded that 19,20-EDP may be an important mediator of monocyte differentiation during fibrosis reversal.
